N1-Methylpseudo-UTP (N1-Methyl-Pseudouridine-5'-Triphosphate) is a modified nucleoside triphosphate designed for incorporation into messenger RNAs (mRNAs) using T7 RNA polymerase. The inclusion of N1-methylpseudouridine reduces the immunogenicity of the resulting mRNA.

A recent study reported that the N1-methylpseudouridine (m1Ψ) modification, either alone or in combination with 5-methylcytidine (m5C), outperformed the current state-of-the-art pseudouridine (Ψ) or m5C/Ψ-modified mRNA platforms. Specifically, it achieved up to ~44-fold higher reporter gene expression in cells (with double-modified mRNAs) and ~13-fold higher expression in mice (with single-modified mRNAs). Furthermore, compared to (m5C/)Ψ-modified mRNAs, those modified with (m5C/)m1Ψ exhibited reduced intracellular innate immunogenicity and improved cellular viability following in vitro transfection. These findings suggest that N1-methylpseudo-UTP may serve as a valuable component for synthesizing druggable mRNAs with enhanced performance.
